Features

The cAMP Maxensor ADORA3 contains HEK293 cells stably expressing cAMP MaxensorFP650 biosensor and adenosine A3 receptor (unlabeled). The Creative Biolabs cAMP Maxensor ADORA3 cell line is designed to test compounds or analyze their ability to modulate adenosine A3 receptors. When the agonist binds to ADORA3, it activates the G protein, which in turn triggers a cellular response mediated by cAMP. This cell line has been verified by measuring the reduction of cAMP in the cytosol and analyzing the distribution of the cAMP Maxensor biosensor in the cell. This cell line allows image analysis of compound-induced stimuli. The use of IB-MECA as an agonist in high-throughput analysis (HTA) validates this highly reproducible assay.

About Red cAMP Maxensor Biosensor:
The red cAMP Maxensor biosensor is a fluorescent peptide that changes its intracellular location in the presence or absence of cAMP. Before stimulating cAMP production, the fluorescent biosensor is located in the cell membrane. The increase in the concentration of the second messenger leads to changes in the structural folding of the red cAMP Maxensor biosensor, which promotes the relocation of cells in the transport of cell vesicles. In cell lines that co-express the red cAMP Maxensor biosensor and the GPCR of interest, the activity of living cells can be easily quantified by fluorescence granularity image analysis or fluorescence intensity analysis.

Assay Description

Adenosine A3 receptor (ADORA3) is a member of the adenosine receptor group of GPCRs and uses adenosine as an endogenous ligand. Activation of the adenosine A3 receptor by an agonist leads to the binding of Gi1/2/3 or Go protein. The binding of Gi1/2/3 leads to the inhibition of adenylate cyclase, therefore, the concentration of cAMP is reduced. Creative Biolabs' Adenosine A3 receptor detection allows the Maxensor biosensor to be used to measure ADORA3 receptor activation through Go activation.
