Description

Black-Gold II is a novel halophosphate complex that localizes myelin sheaths within the central nervous system. The Black Gold II ready-to-use (RTD) staining kit allows you to localize myelin sheaths, including individual fibers and bundles, As well as selection by toluidine blue counterstain co-localized cell bodies Black Gold II-labeled myelinated fibers appear almost black, while toluidine blue O-labeled cells Nissl bodies appear blue under brightfield illumination.

Black Gold II can demonstrate and characterize specific myelin changes associated with exposure to multiple neurotoxicants, including kainic acid, domoic acid, 3-nitropropionic acid, fluorogold, and isoniazid. Black Gold II can also be used in combination with other histochemical markers, including Nissl stain, fluorescent tracers for retrograde transport, and fluorescent markers for neuronal degeneration. The associated advantages of Black-Gold II include high resolution, high contrast, short histochemical processing times, versatility and consistent reproducibility.

Application Notes

Black Gold II is a high resolution myelin stain with an amyloid plaque counter stain. It is suitable for studies using formalin or paraformaldehyde-fixed, non-paraffin-embedded, non-solvent-treated brain tissue. It can be used for thick and thin sections. For thick sections, gelatin-coated slides or slides specifically designed to bind tissue sections should be used to avoid section loss. Free-floating sections are also available, but sections are easier to handle and transfer when mounted on slides. A suggested method for thick sections is provided as a guide: cryosection or vibratome sections are cut at 20-50 µm thickness and collected in 0.1 M neutral phosphate buffer. Sections are then mounted, typically on 1% gel-coated slides, and air-dried on a slide warmer (50 °C) for at least an hour until completely dry and adherent to the slide. Although these sections are easier to handle when mounted on slides, these sections may stain loosely. Fixed sections were rehydrated in distilled water for 2 min before processing in staining solution.
