Product Overview

Description

Brain uptake of therapeutic antibodies is severely limited by their size. To achieve enhanced BBB crossing, Creative Biolabs developed a BBB shuttle antibody platform by utilizing the endogenous macromolecule transportation pathway, known as receptor-mediated transcytosis (RMT). The engineered antibody-based carrier is believed to significantly to increase the macromolecule brain entry to combat CNS diseases.
Notes: The BBB antibody is made-to order and available in a customized format. Please don't hesitate contact us for more details.

FCM

Figure 1 is a series of FACS analyzes demonstrating that anti-CXCR4 scFvs 33 and 48 specifically bind CXCR4. However, these scFvs do not bind the closely related chemokine receptor CCR5.

FCM

Figure 2 is a series of FACS analysis graphs showing that scFvs 48 binds to Cf2ThCD4+CXCR4+ cells but not to parental Cf2Th or Cf2ThCD4+CCR5+ cells.

FCM

Figure 3 is a series of FACS scans showing that scFvs 33 and 48 do not cross-react with other chemokine receptors such as CXCR6, CXCR1, GPR15 and CXCR2.

FCM

Figure 4 is a FACS scan analysis showing that scFv clones 33 and 48 compete with neutralizing anti-CXCR4 monoclonal antibody 12G5 for binding to CXCR4.

FCM

Figure 5 shows the results of FACS analysis of the binding of 33 scFv-Fc and 48 scFv-Fc to 293T cells expressing human, mouse and rhesus monkey CXCR4.

FuncS

Figure 6 is a series of graphs demonstrating inhibition of HIV-1 reporter virus entry into Cf2ThCD4CXCR4 cells.

FuncS

Figure 7 is a series of graphs showing inhibition of HIV-1 reporter virus entry by scFvs 33 and 48.
