Serotonin (5-HT) regulates the activity of almost all cells in the brain and participates in the regulation of behaviors such as emotions, appetite, sleep, and cognition. Imbalance in 5-HT levels is associated with mood disorders such as depression and anxiety. Therefore, real-time and sensitive monitoring of the dynamic changes of 5-HT is crucial to understanding the functions, mechanisms, and neuropsychiatric diseases of the 5-HT system. Traditional methods such as pharmacology, genetics, and calcium probes have the limitation of being unable to directly monitor the dynamic changes of 5-HT in living animals in real time. The new green 5-HT fluorescent probe has greatly improved sensitivity and can more sensitively detect the dynamic changes of 5-HT in living mice. Combined with large field of view imaging, different release characteristics of 5-HT in the cortex were observed, promoting the discovery of new phenomena. The first red 5-HT probe provides a new option for the simultaneous detection of 5-HT and other neurotransmitters. These probes help researchers further understand the physiological functions of 5-HT.
