Product Overview

Description

Brain uptake of therapeutic antibodies is severely limited by their size. To achieve enhanced BBB crossing, Creative Biolabs developed a BBB shuttle antibody platform by utilizing the endogenous macromolecule transportation pathway, known as receptor-mediated transcytosis (RMT). The engineered antibody-based carrier is believed to significantly to increase the macromolecule brain entry to combat CNS diseases.
Notes: The BBB antibody is made-to order and available in a customized format. Please don't hesitate contact us for more details.

Figure 2. Confocal microscopy showing THP-1 cells with monoclonal antibodies.

THP-1 cells were grown to approximately 60% confluency on sterile plastic coverslips. Cells were fixed in acetone for 20 minutes, washed with PBS and incubated with antibodies for 1 hour. Cells were then washed again and incubated with Texas Red anti-mouse IgG for 1 hour in the dark.

FuncS

Figure 3. Shows the effect of monoclonal antibodies on cholesterol efflux from THP-1 cells.

Cholesterol efflux experiments were performed as described in the Examples section. Cholesterol efflux is expressed as the percentage of labeled cholesterol moving from the cells to the medium (i.e., radioactivity in the medium / radioactivity in the medium + radioactivity in the cells). Mean + SD of quadruplicate determinations are shown. *p<0.01 vs. non-specific IgM.

Figure 4. Shows the effect of monoclonal antibodies on cholesterol efflux from THP-1 cells.

Cholesterol efflux experiments were performed as described in the Examples section.

FuncS

Figure 5 shows that antibodies can detect changes in ABCA1 abundance.

RAW 264.7 mouse macrophages were activated by the LXR activator T0901317. Equal amounts of cellular proteins from activated and non-activated cells were analyzed by western blot using antibody NDF4C2 followed by densitometry. A seven-fold increase in the abundance of ABCA1 was detected in RAW 264.7 cells.
