Recombinant NI-202.21D11 is selective for human and mouse α-synuclein. On the other hand, NI-202.12F4 binds human and mouse α-synuclein in direct ELISA.

Recombinant human α-synuclein was coated onto the ELISA plate at the specified concentration, and passed the direct ELISA (□ 20 μg/ml; □ 20 μg/ml;▴ 5 μg/ml; ♦ 1 μg/ml; 0.25 μg/ml;▾0.1 μg/ml). Determine the half-maximal effective concentration (EC50) for each coating concentration, indicating antibody potency.

Pepscan analysis revealed that NI-202.21D11 overlapped with peptides B08 (aa 109-123 of SEQ ID NO: 1), B09 (aa 113-127 of SEQ ID NO: 1) and B10 (aa of SEQ ID NO: 117-131) Binding to human α-synuclein of 1) shows that the minimal sequence required for NI-202.21D11 binding is PVDPDNE within human α-synuclein (aa 117-123 of SEQ ID NO: 1).
