Product Overview

Description

Brain uptake of therapeutic antibodies is severely limited by their size. To achieve enhanced BBB crossing, Creative Biolabs developed a BBB shuttle antibody platform by utilizing the endogenous macromolecule transportation pathway, known as receptor-mediated transcytosis (RMT). The engineered antibody-based carrier is believed to significantly to increase the macromolecule brain entry to combat CNS diseases.
Notes: The BBB antibody is made-to order and available in a customized format. Please don't hesitate contact us for more details.

Product Pictures
FuncS

Figure 1 depicts antibody inhibition of human progranulin binding to human sortin.

FuncS

Figure 2 depicts the level of Sortilin on the cell surface of U-251 cells incubated with different concentrations of anti-Sortilin antibodies for 72 hours.

ELISA

Figure 3 depicts ELISA-based quantification of corresponding levels of PGRN in U-251 cell culture medium

ELISA

Figure 4 depicts the binding of anti-sortilin antibody to sortilin in the presence (dashed line) or absence (solid line) of pro-NGF.

Block

Figure 5 depicts the inhibition of progranulin binding (in percent PGRN blockade) by 150 nM affinity matured anti-sortilin antibody derived from antibody S-15 versus HEK293T cells expressing sortin. The affinity matured antibody S-15-6 showed improvement in PGRN blockade.

FuncS

Figure 6 shows the reduced binding of the S-60 antibody to certain sortin mutants in which human amino acids are replaced with mouse sortin amino acids. The results indicated that 5595 amino acids are required for S-60 binding.

Figure 7 shows the level of binding of the anti-sortilin antibody S-60 to sortilin in the presence or absence of NTS.

WB

Figure 8 depicts the measurement of sorterin levels using Western blot analysis of leukocyte lysates from mice.

FuncS

Figure 9 depicts the results of a collagen-induced arthritis study in mice. Biweekly injection of 40 mg/kg anti-Sortilin antibody S-15-6 significantly reduced clinical arthritis scores compared to controls (PBS and MOPC-21/msIgG1).

FuncS

Figure 10 depicts progranulin (PGRN) in cerebrospinal fluid (CSF) samples obtained from rats two or six days after intra-articular (ia) injection into the knee or intraperitoneal (ip) injection of anti-sortin antibody S-15 ) protein level-6 or control antibody. n=2 rats/group. **p<0.001 One-way ANOVA.
