Features

The red cAMP Maxensor-VIPR2 contains U2OS cells stably expressing the red cAMP Maxensor biosensor and vasoactive intestinal peptide receptor 2 (unlabeled). Creative Biolabs' red cAMP Maxensor-VIPR2 cell line is designed to test compounds or analyze their ability to modulate vasoactive intestinal peptide receptor 2. When the agonist binds to VIPR2, the G protein is activated, which in turn triggers the camp. This cell line has been verified by measuring the increase of cAMP in the cytosol and analyzing the distribution of the red cAMP Maxensor biosensor in the cell. This cell line allows image analysis of compound-induced stimuli. In high-content analysis (HCA) and high-throughput analysis (HTA), the use of human PACAP-38 as an agonist validates this highly reproducible assay.

About Red cAMP Maxensor Biosensor:
The red cAMP Maxensor biosensor is a fluorescent peptide that changes its intracellular location in the presence or absence of cAMP. Before stimulating cAMP production, the fluorescent biosensor is located in the cell membrane. The increase in the concentration of the second messenger leads to changes in the structural folding of the red cAMP Maxensor biosensor, which promotes the relocation of cells in the transport of cell vesicles. In cell lines that co-express the red cAMP Maxensor biosensor and the GPCR of interest, the activity of living cells can be easily quantified by fluorescence granularity image analysis or fluorescence intensity analysis.

Assay Description

The VPAC2 receptor detection (VIPR2) from Creative Biolabs uses Maxensor Biosensors technology to measure VIPR2 activation. VPAC2 receptor signals are stimulated by adenylate cyclase through the trimeric Gs protein, increased cAMP, and downstream L-type calcium channel interactions.

In this assay, we use the cAMP-Maxensor biosensor to measure receptor activation/inhibition through cAMP accumulation. The high reproducibility of this assay allows monitoring of the VPAC2 receptor activation process in living cells.
