The guide strand then precisely scans for and binds to messenger RNA (mRNA) molecules that possess a complementary nucleotide sequence.
- mRNA Cleavage and Gene Silencing: The exquisite sequence complementarity between the siRNA guide strand and the target mRNA triggers the catalytic activity of the RISC, particularly through Argonaute proteins. This leads to the cleavage and subsequent degradation of the mRNA. By preventing the mRNA from being translated into protein, the expression of the corresponding gene is effectively silenced. This high specificity, based on Watson-Crick base pairing, is a hallmark advantage of siRNA.
