Description

AAV-PHP.S is an AAV9 variant engineered through directed evolution of its capsid protein. In mice, it demonstrates over 5 times greater efficiency at penetrating the blood-brain barrier (BBB) compared to AAV9, with a high specificity for neurons of up to 90%. Its infection mechanism relies on sialic acid receptors, but its transduction efficiency in primates still requires optimization.

Whole-Brain Neural Circuit Tracing: After intravenous injection, it enables highly efficient labeling of neurons throughout the entire brain. This can be combined with fluorescent proteins or optogenetic tools to dissect complex neural circuits.
Neurodegenerative Disease Models: It can deliver genes like the amyloid-beta precursor protein (APP) gene to the whole brain, effectively mimicking the pathological progression seen in Alzheimer's disease.
