AD Modeling and Drug Efficacy Testing Service

Disease Mechanism

Alzheimer's disease (AD), also known as senile dementia, is the most common type of senile dementia. The main manifestations are neuropsychiatric symptoms such as progressive memory impairment, cognitive dysfunction, personality changes, and language disorders. The etiology and pathogenesis of AD have not been elucidated. The characteristic pathological changes are extracellular senile plaques formed by β-amyloid deposition and intraneuronal neurofibrillary tangles formed by hyperphosphorylation of tau protein, neuronal loss with glial cell proliferation, etc.

AD Modeling

Classification Modeling Method Animals
Alzheimer's disease model Brain localization surgery Rat
Surgical method Sterile 7mm electrodes from the dura mater and stimulate with 1mA DC Rat
Brain localization injection of amanitamin Rat
Brain-targeted injection of immunotoxin to eliminate basal forebrain cholinergic cells Rat
Intraperitoneal injection of scopolamine Rat
Injection of solution A into the hippocampus Rat
Stereotactic injection of okadaic acid in the brain Rat
Transgenic animals (APP, PS-1, PS-2, and apoE4, etc.) Rat/Mouse
Aluminum trichloride hexahydrate gavage (chronic aluminum poisoning) Rat
D-galactose (D-gal)-induced subacute aging model Rat
24 months old (natural aging) Rat

Assays after Modeling

Creative Biolabs offers a variety of well-accepted behavioral tests to assess potential anxiolytic, antidepressant, or antipsychotic effects, as well as cognitively enhancing and disruptive effects on learning and memory (cognitive, psychiatric, neurological). We have developed a comprehensive range of rat primary neuronal culture models and assays to evaluate the neurotoxic, neuroprotective, or neurotrophic effects of test compounds (cellular models).

1) Cognitive function

2) Imunohistochemical detection (GFAP,NeuN,ChAT,APP,Aβ1-42,p-Tau, etc.), etc.

In addition to the above tests, we can also provide MRI/PET-CT in vivo imaging of small animals, cardiac ultrasound, X-ray films, tissue fluorescence imaging (whole brain slide scan, tissue immunofluorescence imaging), patch clamp on cells and brain slices, multi-channel in vivo electrophysiological recordings, respiration recordings, behavioral testing of pain, anxiety, depression, olfactory function-related (go/no-go) in awake animals.
