Description

The mouse iPS cell line (induced pluripotent stem cell line) is derived from mouse embryonic fibroblasts (MEF) through retroviral expression of Oct3/4, Sox2, Klf4, and c-Myc genes. Morphological selection criteria are used to derive cells. When cultured under standard mouse ES cell culture conditions, the morphology of mouse iPSC is the same as that of mouse ES cells. These cells also expressed the pluripotency markers SSEA-1 and Nanog, and showed strong endogenous alkaline phosphatase activity. Mouse iPS cells grow on the feeder layer of mouse embryonic fibroblasts (MEF), and the plates need to be pretreated with gelatin.

Handling Advice

Frozen cells:Upon receipt, frozen ampoules should be transferred directly to gaseous phase liquid nitrogen without delay, unless they are to be used straight away.

Growing cells: Growing cell cultures should be checked on receipt using an inverted microscope. Immediately check the cell density upon arrival for any obvious defects. If the cell density is too high (more than 80% confluent) subculture the cells (harvest and reseed) immediately.

Warnings

Store in gas phase of liquid nitrogen immediately upon receipt. This product is stable for 6 months when stored as directed.

Quality Control

Mouse iPS cells were grown in mouse ES medium supplemented with 103 U/ml LIF. After recovery from cryopreservation, each batch of mouse iPS cells was tested for growth and viability. In addition, the SSEA-1 and Nanog expression and alkaline phosphatase activity of each batch of cell lines were also tested.
