Features

Each vial contains U2OS cells stably expressing human TAR DNA binding protein 43 (TDP-43) labeled with tGFP. Creative Biolabs' TDP43 translocation detection cell line is designed to detect the ability of compounds or analytical stimuli to modulate TDP-43 translocation, follow the cytoplasmic translocation process and quantify intracellular fluorescence accumulation. This highly reproducible assay allows monitoring of TAR DNA-binding protein 43 translocation in high-content analysis and fluorescence microscopy applications.

TDP-43 Stress Granules Assay

Assay Description

Amyotrophic lateral sclerosis (ALS) is one of the most common degenerative diseases of the motor neuron system. TDP-43 pathology appears to be the main pathological type of sporadic ALS types. Our TDP-43 stress granule assay allows pathological quantification TDP43 spheres enter the nucleus and cytoplasm. This model also allows monitoring the distribution of TDP-43 protein in living cells, studying the protein localization pattern in space and time, and quantifying the intracellular fluorescence aggregation.

For this assay, we use a stable cell line expressing fluorescent TDP-43. During the assay, the cells were incubated with the test compound and sodium arsenite to induce TDP-43 aggregation. Finally, we used BD Pathway HCS Reader and Attovision Compartimentalization software to quantify the TDP43-tGFP nucleosphere.

The cell line has been screened using the luciferase based mycoplasma detection kit to confirm the absence of mycoplasma species.
