Official Full Name: Thyrotropin-releasing hormone receptor
DNA Accession Number: GenBank: AY493373
Host Cell: U2OS
Format: 2 cryopreserved vials
Resistance: Puromycin

Features

The HiTector TRH1 contains U2OS cells stably expressing unlabeled human thyroid-stimulating hormone releasing hormone receptor (TRH1). The HiTector TRH1 cell line is designed to test compounds or analyze their ability to modulate the thyroid-stimulating hormone releasing hormone receptor. When the agonist binds to TRH1, it activates the G protein, which in turn triggers a cellular response mediated by the second messenger (calcium). This cell line has been validated by measuring the increase in calcium in the cytosol. The high reproducibility of this assay allows the TRH1 activation process to be monitored in high-throughput screening.

Assay

Thyrotropin-releasing Hormone Receptor Assay

Assay Description

Thyroid-stimulating hormone-releasing hormone (TRH), also known as thyroid-stimulating hormone-releasing factor (TRF) or thyroid hormone, is one of the smallest hormones. The thyroid-stimulating hormone-releasing hormone receptor assay allows measurement of the activity and potency of recombinant human TRH.

To analyze the activity of rhTRH, we used a stable cell line expressing thyroid stimulating hormone releasing hormone receptor subtype 1 (TRH1). When rhTRH binds to TRH1, it activates the G protein, which in turn triggers a cellular response mediated by the second messenger (Ca2+). The high reproducibility of this assay allows monitoring of the activation process of the thyroid-stimulating hormone releasing hormone receptor in high-throughput screening.

Mycoplasma Testing

The cell line has been screened using the luciferase based mycoplasma detection kit to confirm the absence of mycoplasma species.

Sterility Testing

Sterility testing was performed in accordance with USP and EP regulations. All of our sterility testing is performed in an isolator or clean room environments. The cell line has been screened using the membrane filtration testing methods to confirm the absence of aerobic, anaerobic and fungi microorganisms.

Genetic Stability Testing

Cell genetic stability study was perfomed under ICH guidelines. We provide guidance on the appropriate testing program upon your requirements.
