Description

Cell line information: tGFP-APP MDCK Cell Line (Alzheimer's disease in vitro model)

Features

tGFP-APP MDCK cells allow analysis to assess the endogenous secretase proteolytic process in living cells. This cell line has been validated with different inhibitory compounds and siRNA for BACE to block γ-secretase and β-secretase activities to reduce their processing. Each batch of division-suppressing cells is highly validated and shows the same reference compound response as the stable cell line. The high-content analysis of gamma and beta secretase activity is designed to be performed using an epi-fluorescence imaging system to acquire and analyze images and quantify fluorescent vesicles entering the cytoplasm. The results obtained during the verification of the assay indicate that the pharmacological inhibition of the secretase involved in AD is an effective strategy for drug screening, and these models are suitable for monitoring the disease process in vivo in a bioimaging system.

Assay Description

Creative Biolabs has developed a new fluorescence-based detection method for screening APP Processing Assay inhibitors in the APP Processing pathway. This new detection method requires the use of MDCK cell lines that stably express green fluorescent APP. Treating this cell line with an inhibitor of Aβ peptide formation such as an inhibitor of secretase activity, APP appears to remain in the fluorescent spot. It occurs in a dose response manner, allowing quantification using imaging systems. This model can recognize β-secretase and γ-secretase inhibitors, as well as any other compounds that prevent the formation of Aβ peptides.

Mycoplasma Testing

The cell line has been screened using the luciferase based mycoplasma detection kit to confirm the absence of mycoplasma species.

The cell line has been screened using the membrane filtration testing methods to confirm the absence of aerobic, anaerobic and fungi microorganisms.
