Features

Multiple Tau gene mutations are the causative factors of inherited FTDP-17 disease. These mutations have a similar effect to the hyperphosphorylation of Tau in AD and lead to the formation of NFT. U2OS stably expressing triple mutant (TM) human ON4R Tau-tGFP can be used to evaluate endogenous Tau kinase and phosphatase in living cells. The combination of Tau and MT and continuous bundle formation depend on phosphorylation: hyperphosphorylation of Tau causes it to dissociate from the microtubules and aggregate into tangles of paired spiral filaments (PHF). In contrast, kinase inhibitors promote Tau to bind to microtubules and form microtubule bundles. Automatic image analysis is used to detect and quantify this beam increase through fluorescence. It is suitable for monitoring the disease process in the body in the bioimaging system.

Assay Description

Creative Biolabs' tau phosphorylation assay allows identification of tau kinase and phosphatase inhibitors and modulators that affect the behavior or location of tau protein. In this tau phosphorylation assay, we use a U2OS cell line stably expressing triple mutant (TM) human 0N4R Tau-tGFP. The binding of tau to microtubules and continuous bundle formation depend on phosphorylation. Hyperphosphorylation of tau causes it to dissociate from the microtubules and aggregate into tangles of paired helical filaments (PHF). In contrast, kinase inhibitors promote the binding of tau to microtubules and the formation of microtubule bundles.
