Figure 1 Anti-TREM2 antibody binds to TREM2 on the cell surface.
Anti-TREM2 antibody surface binding dose response in hTREM2-HEK cells. Binding of anti-TREM2 antibodies was titrated to HEK cells expressing human TREM2. Antibody binding was detected with a secondary anti-mouse multiplexed antibody conjugated to APC. RS9.F6 and RS9.F10 demonstrate binding to human TREM2 cells with an EC50 of less than 2 nM.

Determining the P-Syk Activating Dose Response of Anti-TREM2 Antibodies. Anti-TREM2 antibodies were dose titrated to HEK cells expressing human TREM2, and pSyk induction was calculated as a buffer control fold. EC50 values show the nM potency and fold induction of pSyk for each antibody.
Figure 4 Anti-TREM2 antibody induces human macrophage survival.
Human monocytes isolated from peripheral blood were incubated with 5 ng/mL M-CSF in the presence of titrated concentrations of plate-coated anti-TREM2 antibody or an isotype control. On day 6, cell viability was determined by the CellTiter Glo Viability Assay. Anti-TREM2 antibodies increase the survival of human macrophages cultured in limited or no M-CSF.

Human macrophages were stimulated with 30 nM antibody or control for 15 minutes. p-Y525/526-SYK of cell lysates was measured by Alpha-LISA. Anti-TREM2 antibody induces Syk phosphorylation, ERK phosphorylation, GSK3-β phosphorylation and AKT phosphorylation.
