Features

The cAMP Maxensor DRD1 cell line contains HEK293 cells, which stably express red fluorescent cAMP Maxensor biosensor and dopamine receptor D1 (without label). The Creative Biolabs cAMP Maxensor DRD1 cell line is designed to determine compounds or analyze their ability to modulate dopamine receptor D1. When the agonist binds to DRD1, the G protein is activated, which in turn triggers a cellular response mediated by cAMP. This cell line has been verified by measuring the increase of cAMP in the cytoplasm, analyzing the fluorescence intensity of the cAMP Maxensor biosensor and the distribution within the cell. This cell line allows the measurement of the stimulus induced by the test compound by fluorescence intensity and image analysis. This highly reproducible assay has been validated by high-throughput screening (HTS) and high-content analysis (HCA) using dopamine as an agonist.

About Red cAMP Maxensor Biosensor:
The red cAMP Maxensor biosensor is a fluorescent peptide that changes its intracellular location in the presence or absence of cAMP. Before stimulating cAMP production, the fluorescent biosensor is located in the cell membrane. The increase in the concentration of the second messenger leads to changes in the structural folding of the red cAMP Maxensor biosensor, which promotes the relocation of cells in the transport of cell vesicles. In cell lines that co-express the red cAMP Maxensor biosensor and the GPCR of interest, the activity of living cells can be easily quantified by fluorescence granularity image analysis or fluorescence intensity analysis.

Assay Description

Creative Biolabs' D1 dopamine receptor assay allows the measurement of DRD1 receptor activation by the increase in cAMP in the cytosol of the cell. We analyzed the test compounds of the G protein signaling pathway by Gs involving activation of the DRD1 receptor. When the agonist binds to DRD1, it activates the Gs protein; in turn, the cAMP-mediated cellular response is triggered by adenylate cyclase stimulation. This leads to an increase in the fluorescence intensity of the cAMP Maxensor biosensor. Therefore, the activity of living cells can be easily quantified in a dose-response manner through the fluorescence emission in the microplate reader.
