Description

Rat oligodendrocytes are isolated from cerebral cortex tissue; the brain is divided into left and right hemispheres, and the cerebral cortex (gray matter) covers most of each cerebral hemisphere and is where neuronal cell bodies are concentrated. Inside is white matter made up of nerve fibers or myelin sheaths. Each hemisphere has three surfaces, namely the lateral surface (accounting for about 1/3 of the entire cortical area), the medial side and the bottom surface (accounting for 2/3 of the area); the surface of the hemisphere has many grooves or fissures of varying depths. The bulges between the fissures are called gyri, and they greatly increase the surface area of ​​the brain; the important grooves and fissures on the lateral side of the brain include the Sylvian fissure, the parieto-occipital fissure, and the central fissure. The cerebral cortex is divided into four major parts: frontal lobe, parietal lobe, temporal lobe, and occipital lobe due to the boundary between the three fissures. Oligodendrocytes are distributed in the central nervous system. In silver-stained specimens, oligodendrocytes are smaller than astrocytes, and their protrusions are also smaller and less, in the shape of beads, so they are called oligodendrocytes. cells or oligodendrocytes. However, the oligodendrocytes shown by specific immunocytochemical staining have not many protrusions and many branches. The main function of oligodendrocytes is to wrap axons in the central nervous system, form an insulating myelin sheath, assist in the efficient transmission of bioelectrical signals, and maintain and protect the normal function of neurons. Its abnormality will not only lead to demyelinating lesions of the central nervous system, but also cause neuronal damage or psychiatric diseases, and even brain tumors. According to the distribution and location of oligodendrocytes, they can be divided into three types: ① interfascicular-oligodendrocytes, which are distributed between the nerve fiber bundles in the white matter of the central nervous system; ② perineural oligodendrocytes Glial cells (perineuronal-oligodendrocyte), distributed in the gray matter area of ​​the central nervous system, are often located around nerve cells and are closely related to nerve cells, so they are also called perineuronal-satellite-cells. There are also often lamellar processes of astrocytes between the nerve cell body and such cells; ③ perivascular oligodendrocytes (pcrivascular-oligodendrocytes), mainly distributed around the blood vessels in the central nervous system. The myelin membrane formed by oligodendrocytes is one of the most specialized and complex animal cell membranes, and there are many transmembrane proteins and surface proteins on it to maintain the dense and stable structure of myelin. Such as galactocerebroside (GC), which is a major lipid of myelin sheath, the identification of mature oligodendrocytes with GC antibodies is an early immunological identification method. In the past decade, the field of neurodevelopment has progressed rapidly, and more oligodendrocyte molecular markers have been identified, such as PDGFRa, Mbp, CNP, and SOX-10.
