Description

Sirtuins are a class of proteins with histone deacetylase or monoribosyl transferase activity. Sirtuins are located in the cytoplasm, nucleus, nucleolus and mitochondria. They are related to aging, cell protection, sugar metabolism and cell cycle regulation. Unlike other known protein deacetylases that simply hydrolyze acetyllysine residues, sirtuin-mediated deacetylation reaction hydrolyzes acetyllysine and NAD. This hydrolysis produces deacetylated substrates, O-acetyl-ADP-ribose and nicotinamide, which themselves are inhibitors of sirtuin activity. Studies have shown that human sirtuins may have intracellular regulatory proteins with single ADP-ribosyltransferase activity. In our Sirtuin activity detection kit, the acetylated p53-AFC substrate is deacetylated by Sirtuins in the presence of NAD + to generate deacetylated p53-AFC substrate, nicotinamide and O-acetyl-ADP ribose. The cleavage of the deacetylated p53-AFC substrate by the imaging agent releases a fluorophore, which is fluorescently detected at Ex / Em = 400/505 nm. HDAC can also deacetylate the acetylated p53-AFC substrate. Trichostatin A was added to the reaction to specifically inhibit HDAC in the sample. The kit provides a fast, simple, sensitive and reliable test method that can measure Sirtuin activity in various samples. The limit of quantification of this analysis is 0.06 µU recombinant human SIRT6.
