Description

Human Osteopontin (OPN) is a negatively charged 314 amino acid hydrophilic protein and undergoes important post-translational modifications (PTM), including phosphorylation and glycosylation. Due to its acidic and PTM properties, OPN works abnormally through SDS-PAGE. Its mass is 35kD, and its apparent molecular weight may be as high as 75kD. In addition, OPN undergoes proteolytic modification into smaller molecular weight fragments. The stromal cell protein osteopontin binds to cell surface receptors and is secreted into many body fluids, including milk, blood and urine, according to the organ of origin. This makes osteopontin an ideal candidate for biomarkers because it is easy to obtain a secreted form in the waste liquid and mimics the cellular environment from which it is released. Osteopontin is important in immune response and inflammation, as well as bone production and remodeling. In children with autism, the serum level of osteopontin correlates with the severity of the disease, and may be due to the pattern of inflammation in the brain of these children. In aortic valve sclerosis and stenosis, an increase in the level of secreted osteopontin is also noted. Osteopontin is also considered a biomarker of cancer because it is related to tumor formation, progression and metastasis. During the formation of bones and teeth, osteopontin is a negative regulator of parathyroid hormone-related protein receptors and can induce bone formation. Without proper levels of osteopontin, bone growth will continue to be uncontrolled and lead to specific bone cancers. In short, osteopontin is a powerful marker of bone growth, inflammation and certain cancers.
