Target Overview

Apolipoprotein E (APOE) is a central protein in lipid metabolism, primarily responsible for transporting cholesterol and other fats. There are three major versions (alleles) of the APOE gene: APOE2, APOE3, and APOE4. The APOE4 isoform is the single most significant genetic risk factor for developing late-onset Alzheimer's disease. Compared to the more common APOE3 form, the APOE4 protein is associated with increased aggregation of amyloid-beta plaques and the formation of tau tangles, both of which are hallmarks of Alzheimer's pathology. This makes the APOE4 isoform a critical target for diagnostic and therapeutic research.
