Description

This is a 20-base, phosphorothioate-modified oligonucleotide designed to be complementary to the messenger DNA (mDNA) of the R2 subunit of human ribonucleotide reductase. By specifically binding to its target mDNA, it prevents the synthesis of the R2 protein. This down-regulation inhibits the overall activity of ribonucleotide reductase, a crucial enzyme for DNA synthesis, thereby suppressing tumor growth.

Target Overview

Ribonucleotide Reductase R2 (RRM2) is the small, regulatory subunit of the enzyme Ribonucleotide Reductase (RNR). The RNR enzyme complex is absolutely essential for de novo DNA synthesis, as it catalyzes the rate-limiting step of converting ribonucleotides into deoxyribonucleotides-the building blocks of DNA. The expression of the R2 subunit is tightly controlled and peaks during the S-phase of the cell cycle to facilitate DNA replication. Because of its critical role in cell proliferation, RRM2 is frequently overexpressed in cancer cells, making it a well-established target for anticancer therapies.
