Description

This is a compact, single-stranded DNA aptamer composed of 15 deoxynucleotides that folds into a stable G-quadruplex secondary structure. This well-defined conformation enables it to bind with high affinity and specificity to the key coagulation enzyme, thrombin. By occupying the active site or an allosteric site, the aptamer directly blocks thrombin's proteolytic function, thereby preventing the conversion of fibrinogen into fibrin, which is the final and essential step in blood clot formation.

Target Overview

Thrombin is the central effector serine protease of the blood coagulation cascade. It is generated from its inactive zymogen, prothrombin, during the final steps of the cascade. Thrombin's primary role is to convert soluble fibrinogen into insoluble fibrin strands, which form the meshwork of a blood clot. Beyond this, it is a powerful prothrombotic agent that amplifies its own production by activating other coagulation factors (V, VIII, XI) and platelets. Due to its pivotal and multifunctional role in both hemostasis and pathological thrombosis, thrombin is a principal target for many anticoagulant therapies.
