Glutaminase (EC 3.5.1.2, GLS) is an enzyme that hydrolyzes glutamine and produces glutamate and ammonia. It has tissue-specific effects in multiple organs, some of which include the production of ammonia in the liver to synthesize urea. When kidney acidosis occurs in the kidney, it maintains acid-base homeostasis through the production of ammonia; and regulates the neurotransmitter glutamate in the brain. Isoglutaminase 1 (GLS1) or phosphoric acid-activated mitochondrial glutaminase is very important for cancer cells to utilize glutamine. It is well known that the rate of glutamine decomposition increases in tumors, so it is harmful to tumor cells. Metabolism is extremely important. GLS1 inhibitors are considered as potential cancer treatment candidates. Our glutaminase (GLS1) inhibitor screening kit is a plate-based fluorescence detection kit for screening human GLS1 inhibitors. Glutaminase hydrolyzes glutamine to form glutamate and ammonia. In the presence of a mixture of developer and enzyme, glutamate converts the non-fluorescent probe into a fluorescent product through an enzymatic reaction. The well-known glutaminase inhibitor-CB-839 is provided as an inhibitor control.
