Description

Proteasomes are very large protein assemblies (20S, 26S) found in the nucleus and cytoplasm of all eukaryotes (and some prokaryotes). They are responsible for the degradation and recovery of previously labeled proteins with ubiquitin. This labeled protein is degraded into peptides about 7-8 amino acids long, which are then further degraded. The 20S assembly is a functional protease structure with chymotrypsin-like, trypsin-like and caspase-like protease activities. Our proteasome activity assay utilizes chymotrypsin-like activity and utilizes an AMC-tagged peptide substrate that releases free, highly fluorescent AMC in the presence of proteolytic activity.
