Description

Apoptosis-related punctate protein (CSC) (ASC), also known as methylation-mediated silencing (TMS1), is the target of PYD and CARD domain-containing protein (PYCARD), and is a 21.6 kD pro-apoptosis Death protein, which contains the N-terminal pyridine domain (PYD) and the C-terminal caspase recruitment domain (CARD). It was initially found that the TMS1 gene was abnormally methylated and silenced in various cancer cells. The expression of ASC can be induced by pro-apoptotic/inflammatory stimuli. In normal cells, proteins are located in the cytoplasm. However, in cells undergoing apoptosis, it forms spherical aggregates near the periphery of the nucleus. In the process of apoptosis, ASC transfers from the cytosol to the mitochondria and binds to the mitochondria Bax, which triggers the release of cytochrome c and subsequent apoptosis. The adaptor molecule ASC mediates AIM2-dependent caspase-1 activation. In addition, the AIM2/ASC complex can act as a new type of caspase-8 activation platform. Recent studies have shown that ASC reduces the phosphorylation of IKKα/β phosphorylation and inhibits the activity of NF-κB in primary melanoma. However, in metastatic melanoma, it has the opposite function of enhancing NF-κB activity and IL-1β secretion.
