SHANK3 is a large scaffolding protein located at the postsynaptic density (PSD) of excitatory synapses within the brain. It functions as a master organizer by anchoring and connecting neurotransmitter receptors, ion channels, and signaling proteins to the underlying actin cytoskeleton. This structural role is essential for the proper development, maturation, and plasticity of synapses. Disruption of the SHANK3 gene through mutation or deletion is one of the most common single-gene causes of Autism Spectrum Disorder (ASD) and is the defining genetic feature of Phelan-McDermid Syndrome, highlighting its critical role in brain development.
