Description

In the early 1990s, an unusual protocol resulted in the production of a mouse monoclonal antibody, clone A60, against a component of the neuronal nucleus and proximal nuclear membrane (1). Therefore, this component was named "NeuN" and showed two protein bands corresponding to 46 and 48 kDa on the Western blot. This antibody is widely used as a reliable neuronal marker and apparently binds to neurons in all vertebrates, although some neuronal cell types are not recognized, such as cerebellar Purkinje cells, olfactory mitral valve cells, and many types of retina Neuron, NeuN immunoreactivity has been recognized and widely used to recognize neurons. Until 2009, Kim and others knew the identity of the NeuN protein. (2) It is shown that it is the same as FOX3, which is the mammalian homologue of the gene product originally identified and named FOX1 in C. elegans (2,3). There are three mammalian homologs, FOX1, FOX2, and FOX3, which are believed to play a role in the regulation of mRNA splicing (4). All three contain an almost identical central RNA recognition motif or RRM domain, which is a region of about 90 amino acids found in many proteins, and specifically binds hexaribonucleotides in all three molecules UGCAUG (4). The four protein isoforms of FOX3 are derived from the alternate splicing of two exons of a single gene, which encode inserts near the C-terminus and short C-terminus extensions (5). This extension includes a C-terminal proline-tyrosine sequence, preceded by a hydrophobic amino acid (Φ-PY), which is known to play a role in nuclear localization, apparently explaining that FOX3 is present in the nucleus and cytoplasm of certain neurons in.
