Description

The mCherry protein is derived from the natural product DsRed, which was originally isolated as red fluorescent protein from corals of the genus Discosoma (1). Like other natural fluorescent proteins of cnidarians (jellyfish, sea anemones, and corals), the natural form of the protein forms a stable tetramer in the body. In Roger Tsien's laboratory, DsRed was designed to improve its spectral characteristics and prevent multimerization, where a lot of work on fluorescent proteins has been done (2). Further cycles of mutation, directed modification, and evolutionary selection produced mCherry, which is a monomer with maximum excitation at 587nm and maximum emission at 610nm (3).
