Target Overview

Disrupted in Schizophrenia 1 (DISC1) is a scaffolding protein that plays a critical role in the brain by coordinating a wide range of signaling pathways essential for neurodevelopment. Its functions include regulating neuronal proliferation, differentiation, migration, and synaptic integration. The DISC1 gene was famously identified through its disruption by a chromosomal translocation in a Scottish family with a high incidence of major mental illness. It has since become one of the most significant and widely studied genetic risk factors for a spectrum of severe psychiatric disorders, including schizophrenia, bipolar disorder, and major depression.
