Description

This is a 26-base, guanine-rich single-stranded DNA aptamer that self-assembles into a highly stable G-quadruplex structure. It acts as an anticancer agent by binding with high affinity and specificity to nucleolin, a protein that is aberrantly overexpressed on the surface of many types of cancer cells. This binding disrupts the multiple oncogenic functions of nucleolin, leading to the inhibition of cancer cell proliferation and the induction of apoptosis.

Target Overview

Nucleolin (NCL) is a highly abundant, multifunctional phosphoprotein found primarily in the nucleolus of growing eukaryotic cells. It is critically involved in multiple stages of ribosome biogenesis, including the transcription of ribosomal DNA (rDNA), maturation and processing of pre-rRNA, and the assembly of ribosomal subunits. Beyond the nucleolus, Nucleolin shuttles to the cytoplasm and can also be found on the cell surface, where it acts as a receptor for various ligands. Due to its overexpression on the surface of cancer cells and its central role in cell proliferation, survival, and growth, Nucleolin has become a key therapeutic target in oncology.
