Target Overview

Parkin is a crucial E3 ubiquitin ligase, an enzyme that attaches ubiquitin tags to other proteins to mark them for degradation by the cell's proteasome system. It plays a vital role in cellular quality control, most notably in a process called mitophagy, which is the selective clearance of damaged or dysfunctional mitochondria. Loss-of-function mutations in the PARK2 gene, which codes for the Parkin protein, are the most common cause of autosomal recessive, early-onset Parkinson's disease. The resulting failure of the mitophagy pathway is thought to lead to the accumulation of toxic, damaged mitochondria and subsequent death of dopamine-producing neurons.
