Description

Rat scleral fibroblasts were isolated from sclera tissue, one of the main components of the eyeball wall. It is the posterior 5/6 of the fibrous membrane of the eye. Connects to the cornea anteriorly, and extends posteriorly to the sheath of the optic nerve. The sclera is thickest near the exit of the optic nerve, becomes thinner as it progresses, and thickens again at the attachment of the tendon. At the junction with the cornea, there is an annular corneal groove on the outside and a canal of the sclera in the deep. The sclera is light blue in children, white in adults, and yellowish in old age due to fat deposition. The sclera is a tough structure that supports and protects the tissues in the eye. Fibroblasts are the main cellular components of loose connective tissue, differentiated from mesenchymal cells in the embryonic stage; fibroblasts are large, with clear outlines, mostly protruding spindle-shaped or star-shaped flat structures, and their nuclei are Regularly oval, with large and prominent nucleoli. Fibroblasts have vigorous functional activities, weak basophilic cytoplasm, and obvious protein synthesis and secretion activities. Under certain conditions, they can achieve mutual transformation with fibroblasts; fibroblasts are resistant to different degrees of cell degeneration, necrosis and tissue. Defect repair plays a very important role. Freshly isolated fibroblasts are round, have good refractive index, and are suspended in the medium. After 30 minutes, the cells adhered to the wall, and some of them began to protrude pseudopodia, which appeared as small protrusions; after 6 hours, the cells were basically completely adherent, stretched into a spindle shape, and the nuclei were clear and evenly distributed. Rapidly, 5-7 days, the cells are in a confluent state, the cells are closely arranged, and some overlap and grow, flat, with large cell bodies, transparent cytoplasm, and large nuclei, which are oval and light in color. Cells fuse and connect to each other in a network; cells are flattened in the shape of a protruding spindle or star.
