Description

Transglutaminase (EC 2.3.2.13) is a calcium-dependent enzyme that catalyzes the post-translational modification of proteins by forming isopeptide bonds. This occurs either by cross-linking of proteins that form γ-glutamyl-ε-lysine bonds, or by introducing primary amines on the glutamine residues to which selected peptides are bound. The transglutaminase family includes most of the intracellular forms (TG1, TG3 and TG5) expressed in epithelial tissues. TG2 is expressed inside and outside the cell and in various tissue types; TG4 is expressed in the prostate; factor XIII is expressed in the blood; TG6 and TG7, whose tissue distribution is unknown, and there is a band 4.2 on the red blood cell membrane (lack Enzymatic activity). Transglutaminase also exhibits GTPase, phosphodiesterase and protein kinase activities. Transglutaminase is related to certain nervous system diseases, autoimmune diseases and cancer. Our transglutaminase activity detection kit utilizes the deamidation reaction of transglutaminase with donor and acceptor substrates to form hydroxamate products. The hydroxamate product reacts with the stop solution to form a purple complex, which can be colorimetrically determined at 525 nm. The limit of quantification for this assay is ~10 µU or 80 ng recombinant hTG2 enzyme.
