Description
Calpain 1 is a calcium-dependent, non-lysosomal cysteine protease with low calcium requirements. It is expressed ubiquitously in the brain, mainly in the cell bodies and dendrites of neurons, and has a low degree of expression in axons and glial cells. Excessive activation of calpain is related to many pathologies related to changes in calcium homeostasis, such as Alzheimer's disease and secondary brain (neuron) ischemia, traumatic brain injury and spinal cord injury caused by acute cellular stress transsexual. Analyze Genie's human calpain 1 inhibitor screening kit based on the detection of the cleavage of the calpain substrate Ac-LLY-AFC. Ac-LLY-AFC has very weak fluorescence; after cleavage of the substrate by calpain 1, free AFC can be quantified by fluorescence (Ex / Em = 400/505 nm). In the presence of strong calpain 1 inhibitors, the hydrolysis of the substrate will be hindered. The kit provides a fast, simple, sensitive and reliable test method, which is suitable for high-throughput screening of Calpain 1 inhibitors. Inhibitor control (calpain inhibitor 1, ALLN) is included to compare the efficacy of test inhibitors.
