Tau protein is a low-molecular-weight microtubule-associated protein (MAP) identified while investigating components essential for microtubule formation. It is extremely soluble and can be detected in neurons, oligodendrocytes, and astrocytes in both the central and peripheral nervous systems. Tau protein mainly exists in axons and is involved in regulating the polymerization and stability of microtubules. Tau, also known as microtubule-associated protein tau (MAPT), is associated with the pathogenesis of several neurological diseases, including Alzheimer's disease (AD), Autism, and Epilepsy. The Function of Tau Protein in AD

Neurofibrillary tangles (NFT) in AD are induced by tau hyperphosphorylation, which occurs at multiple sites, often by the kinases GSK3 and CDK5, and leads to a reduction in the ability of tau protein to bind to microtubules. Hyperphosphorylation leads to tau dissociation and subsequent microtubule instability. Tau oligomerization also occurs. The NFT observed in AD and other tau protein diseases are aggregates of paired helical filaments composed of hyperphosphorylated tau.

One hypothesis indicates that "prion-like self-replication and reproduction" is the mechanism underlying tau aggregation and proliferation in tau protein disorders. Precursor fibrils are formed when tau "seeds" or fibers are moved from a donor cell to a recipient cell. The recipient cell subsequently aggregates additional endogenous tau protein.
