Description

D-amino acid oxidase (DAAO, DAO, EC 1.4.3.3) is a flavoprotein that oxidizes D-amino acids to form hydrogen peroxide and corresponding imino acids. In the second catalytic step of the same enzyme, they are Hydrolysis to α-keto acid and ammonia. In recent years, the possible role of DAAO in schizophrenia has been studied. The simultaneous expression of DAAO and the putative product from the G72 gene may increase the risk of schizophrenia in patients. For example, compared with healthy controls, DAAO expression and enzyme activity were increased in post-mortem brain tissue samples from patients with schizophrenia. The expression of DAAO in the human brain is very special: it can only be found in the cerebellum, but cannot be detected in any other part of the brain. DAAO regulates the physiological levels of D-amino acids such as D-serine, D-aspartic acid and D-alanine. D-serine can act as a modulator of N-methyl-D-aspartate receptors (NMDA receptors, NMDAR), while D-aspartic acid and D-alanine can affect Alzheimer's The white matter and gray matter in the brain of patients with the disease are both elevated. In this assay, DAAO converts D-amino acids as a by-product into hydrogen peroxide. Therefore, the formed by-products will react with the probe and generate a strong fluorescent signal (Ex / Em = 535 / 587nm). The kit is simple, sensitive, high-throughput adaptable, and can detect DAAO activity as low as 0.5 µU.
