Description

The γ-secretase activity assay cell line is an in vitro model of Alzheimer's disease used to identify inhibitors of γ-secretase activity. It contains a U2OS cell line stably expressing the green fluorescent APP-C99 construct, which contains a γ-secretase cleavage site. For testing, cells were incubated with test compounds for 24 hours to evaluate their effect on APP processing. After incubation, the nucleus can be stained with DAPI and the retained fluorescent APP vesicles can be detected by fluorescence using image analysis algorithms.

Features

Each vial of the U2OS cell line stably expressing human tGFP-APP-C99 contains 3 million cells, and Creative Biolabs provides 2 vials of cells for each order. According to the cell culture manual, these cells can be guaranteed to pass more than 30 generations.

Mycoplasma Testing

The cell line has been screened using the luciferase based mycoplasma detection kit to confirm the absence of mycoplasma species.
