Description

Glutamate dehydrogenase (GDH) (1.4.1.2) is a hexamerase that catalyzes the reversible conversion of glutamate to α-ketoglutarate and ammonia, and at the same time reduces NAD + to NADH. Glutamate can be converted into α-ketoglutarate through GDH, and then flow into the TCA cycle, where it can be used to support oxidative phosphorylation, lipid production or supplement key intermediates (such as oxaloacetic acid). It is a key enzyme in the metabolic pathway of glutamine. Overexpression of GDH has been shown to promote cell proliferation, migration and invasion in a variety of tumors, such as colorectal cancer. The increased dependence on glutamine metabolism makes GDH a target for cancer therapeutic intervention. GDH inhibitors have been shown to be toxic to cancer cells in vitro. In our glutamate dehydrogenase inhibitor screening kit, glutamate is deaminated by NADH that produces GDH, which converts non-fluorescent probes into fluorescent products in the presence of an enzyme mixture, and the mixture passes fluorescence Detection is at Ex / Em = 535/587 nm.
