Target Overview

Progranulin (GRN) is a secreted growth factor involved in a variety of cellular processes, including cell proliferation, wound healing, and inflammation. In the central nervous system, it also plays a critical role in lysosomal function and neuronal survival. The importance of Progranulin in brain health is underscored by the fact that heterozygous loss-of-function mutations in the GRN gene are a major cause of inherited Frontotemporal Dementia (FTD), a progressive neurodegenerative disorder. This deficiency, known as progranulin haploinsufficiency, leads to lysosomal dysfunction and is often associated with the pathological aggregation of TDP-43 protein, marking GRN as a key therapeutic target for FTD.
