Histone deacetylase (HDAC) plays a central role in controlling cell cycle regulation, cell differentiation and tissue development. These proteins play a vital role in development and physiology, especially in the heart and nervous system. They are also deeply involved in cell proliferation, cell cycle and apoptosis. HDAC3 is mainly located in the nucleus, but can also be present in the cytoplasm and plasma membrane. The HDAC3 IP and activity assay kit provides an antibody-based method that can specifically immunoprecipitate HDAC3 complexes from cells and tissues and measure HDAC3 activity by fluorescence. Use HDAC3 antibody to immunoprecipitate HDA3 from cell or nuclear extracts, and then capture the complex on protein A/G magnetic beads. The immunoprecipitated complex is incubated with the HDAC substrate. The developer only cleaves the deacetylated substrate to produce a fluorophore, which can be easily analyzed using a fluorescence microplate reader.
