Description

Monoacylglycerol lipase (MAGL, E.C.3.1.1.23) is an important enzyme that regulates endocannabinoid signaling in human physiology. MAGL is a serine hydrolase that can generate free fatty acids and glycerol from monoacylglycerol storage. A free fatty acid often produced by MAGL activity, arachidonic acid is the precursor of eicosanoids, which are a family of pro-inflammatory signaling molecules. The glycerol part can also be used as the basis of metabolism and energy source for various purposes. Inhibition or inactivation of MAGL leads to accumulation of 2-arachidonic acid glycerides (2-AG), which has numerous biological consequences. Desensitization of endocannabinoid receptors and reduction of addiction withdrawal symptoms are one of the results of MAGL inhibition. In addition, the downstream eicosanoid signal pathway is affected; this pathway plays a role in a variety of physiological processes, including inflammation, immunity, nociception, and blood pressure. As the central node that regulates lipid signaling and energy mobilization, MAGL has attracted great interest as a therapeutic target. Our monoacylglycerol lipase inhibitor screening kit provides a direct method to identify inhibitors of human MAGL. In this assay, human recombinant MAGL is used to cleave the MAGL substrate, resulting in increased fluorescence, which can be eliminated by inhibitors of this enzyme. Due to the important role of MAGL in lipid metabolism and energy storage, specific inhibitors of this enzyme have important clinical significance.
