Glutamate decarboxylase 2 is a 65 kDa isoform of glutamate decarboxylase encoded by the GAD2 gene and mainly expressed by the nerve terminal, as opposed to the 67 kDa isoform (GAD67) encoded by the GAD1 gene mainly in the cell body. Autoantibodies against glutamate decarboxylase 2 have been detected in autoimmune diseases including Graves disease (Kallmann et al. 1997) and stiffness syndrome (Raju et al. 2005), and in patients with type I diabetes or high-risk populations Type I diabetes is the most common (Petersen et al., 1994). The N-terminal region of GAD65 is essential for targeting enzymes to secretory vesicles containing GABA (Shi et al., 1994).
