Description

Neurofilament is a 10nm or intermediate filament protein that is specifically found in neurons. It is mainly composed of three main proteins called NF-L, NF-M and NF-H, but other proteins may also exist. NF-H is a neurofilament high or heavy molecular weight peptide. It runs on 200-220 kDa SDS-PAGE gels and has some variability across species boundaries. In fact, the molecular size of the protein is much smaller, about 110kDa (1,2). The unusual SDS-PAGE mobility is partly due to the very high content of charged amino acids, especially the glutamate-rich regions, where the non-phosphorylated form runs at approximately 160 kDa on SDS-PAGE. The main type of NF-H is the axon form, which is heavily phosphorylated on 40 or more tandem repeats containing Lysine-Serine-Proline (KSP) peptides (3-5). The phosphorylation of these peptides caused a considerable further delay on the SDS-PAGE gel, so the highly phosphorylated axon form runs at 200-220kDa with some species variability. NF-H antibodies can be used to identify axon processes in tissue sections and cultures. NF-H antibodies can also be used to visualize neurofilament accumulation seen in many neurological diseases, such as amyotrophic lateral sclerosis (also known as Lou Gehrig's disease), Alzheimer's disease, and post-traumatic injury. The phosphorylated axon form of NF-H, commonly referred to as pNF-H, can be detected in the blood and CSF after various injuries and disease states that cause axon damage, and antibodies such as these can be used to quantify such persistence. Loss of axons (e.g. 6-8).
