glutamic acid decarboxylase (GAD)

Alternative Names

Glutamate decarboxylase or glutamate decarboxylase (GAD) is an enzyme that catalyzes the decarboxylation of glutamate into GABA and CO 2. GAD uses PLP as a cofactor. The reaction proceeds as follows:
HOOC-CH2-CH2-CH(NH2)-COOH→CO2 + HOOC-CH2-CH2-CH2NH2

In mammals; GAD exists in two isoforms with molecular weights of 67 and 65 kDa (GAD67 and GAD65); which are encoded by two different genes on different chromosomes (GAD1 and GAD2 genes; which are human 2 and Chromosome 10). GAD67 and GAD65 are expressed in the brain using GABA as a neurotransmitter; and are also expressed in insulin-producing beta cells of the pancreas; depending on the species. These two enzymes together maintain the main physiological supply of mammalian GABA; although it may also be caused by the enteric nervous system; brain; and other places through the action of diamines Synthesized from putrescine. Oxidase and aldehyde dehydrogenase 1a1.

Several truncated transcripts and peptides of GAD67 can be detected in the developing brain; but their function (if any) is unknown.
