Description

Phenylalanine aminolyase (PAL, EC 4.3.1.24) catalyzes the reversible non-oxidative deamination reaction of L-phenylalanine to generate trans-cinnamic acid and ammonia without any external cofactors. PAL is expressed in plants, yeast and some fungi, but does not exist in bacteria and animals. In patients with genetic diseases, phenylketonuria cannot metabolize phenylalanine, leading to irreversible cognitive impairment. Historically, patients with phenylketonuria have been treated with a restricted diet of phenylalanine throughout their lives. Recently, drugs containing PEGylated PAL have been approved by the FDA for the treatment of phenylketonuria, thereby restricting dietary restrictions. Our phenylalanine aminolyase activity assay kit measures PAL activity through a two-step plate-based assay. The PAL enzymatic reaction product produced in the first step reacts with the imaging agent, resulting in the formation of a fluorescent compound, which is detected at Ex / Em = 410/470 nm. Under the detection conditions, the kit can detect PAL as low as 3 µUnit (3 pmol substrate catalyzed per minute).
