Description

Soluble epoxy hydrolase or sEH (EC 3.3.2.10) is a cytoplasmic enzyme that is ubiquitous in many organs, including liver, kidney, pancreatic islets, pituitary, lymphatic tissue, muscle and gastrointestinal tract. It catalyzes the hydrolysis of epoxyeicosatrienoic acid (EET), an epoxide derived from cytochrome P-450-mediated arachidonic acid metabolism, and forms adjacent diols. sEH plays a major role in the metabolism of endogenous lipids related to pain and inflammation. The soluble epoxy hydrolase activity assay kit is a microplate-based fluorescence assay kit for measuring sEH activity and purified protein in cells and tissues. It is based on the ability of sEH to hydrolyze non-fluorescent substrates into fluorescent products. The kit includes specific inhibitors for soluble epoxide hydrolase, because the substrate can be hydrolyzed by non-specific hydrolase present in cell and tissue lysates. By subtracting the activity of the presence of the sEH inhibitor from the total activity, the specific sEH activity can be obtained.
