Product Overview

Description

A comprehensive kit for the potent knockdown of the Leucine-rich repeat kinase 2 (LRRK2) gene, the most common genetic cause of Parkinson's disease. The set includes three distinct shRNA plasmid vectors targeting LRRK2 mRNA for robust silencing, along with a non-targeting scrambled shRNA plasmid as a negative control. This is a critical tool for studying the pathological effects of LRRK2 kinase activity and validating it as a therapeutic target.

Target Overview

Leucine-rich repeat kinase 2 (LRRK2) is a large, multi-domain enzyme that has both kinase and GTPase enzymatic functions. While its precise physiological roles are still being fully elucidated, LRRK2 is best known for its strong genetic association with Parkinson's disease. Mutations in the LRRK2 gene are the most frequent cause of both familial (inherited) and sporadic Parkinson's disease. These pathogenic mutations typically lead to a toxic gain-of-function by increasing the protein's kinase activity, which is thought to contribute to neuronal damage. This makes LRRK2 a primary therapeutic target for the development of kinase inhibitors.
