Description

The mCherry protein is derived from the natural product DsRed, which was originally isolated as red fluorescent protein from corals of the genus Discosoma (1). Like other natural fluorescent proteins in cnidarians (jellyfish, sea anemones, and corals), the natural form of this protein forms a stable tetramer in the body. In the Tsien laboratory, DsRed aims to improve its spectral properties and prevent multimerization, where a lot of work on fluorescent proteins has been done (2). Roger Tsien, Martin Chalfie and Osamyu Shinomura won the 2008 Nobel Prize in Chemistry for the discovery and development of cnidarian fluorescent protein. Several further cycles of mutation, directed modification, and evolutionary selection produced mCherry, which is a monomer with maximum excitation at 587nm and maximum emission at 610nm (3). The protein is widely used as a fluorescent tracer in transfection, genetic modification, photobleaching, and FRET type experiments. The prototype of these fluorescent proteins is Green Fluorescent Protein (GFP), which is a protein of approximately 27 kDa, originally isolated from the jellyfish Aequoria victoria (4). The mCherry protein is similar to GFP (5,6) in size and general structural properties, but apparently produces a red rather than green fluorescent dye. Like GFP, mCherry only needs molecular oxygen to become fluorescent due to its intrinsic properties, so it can be easily expressed in various systems.
